I have read that taking 2-3 years off from Botox can make it effective again. Does anyone have experience with a resistant patient seeing results after taking time off from injections?

The wrinkle you are seeing is likely from the surrounding area being smoothed by the Botox. It should gradually return to normal as the effects of the Botox wear off. Typically this takes 3 to 4 months.
It is not common to have a lump develop days after Botox treatments. I recommend a visit to your injector for an examination. At times minor bumps from injection occur, but they typically resolve over the first few hours.
Yes it is fine to have the same area injected with both Dysport and Botox. Be sure to wait the full two weeks between the treatments. It takes two weeks for the final results and the injector will need to assess those in order to select the proper number of units for the touch up.
